Development and testing of PHYA gene-specific primers in sorghum to detect light-tolerant traits

Abstract

The PHYA gene s one of the genes that play a role n the shade avodance mechansm. The study amed to desgn specfc prmers for the PHYA gene n sorghum plants and optmze the DNA amplfcaton process. Prmers were desgned usng the Prmer3 Plus onlne tool, and prmers were generated and tested on 14 Indonesan sorghum varetes. Gene amplfcaton wth PCR technology requres PHYA-specfc prmers. The amplfcaton rate reached 100% wth anelng temperatures rangng from 54-55°C specfc to each prmer. The resultng band was brght, frm, and specfc, and the amplcon sze was on target (± 900 bp). The resultng prmers can be used to amplfy the PHYA gene n sorghum plants, whch are then used for the sequencng process so that the base sequence of the PHYA gene n sorghum plants can be known.
